Emergency services were called to Powdermill Lane in Battle, near Hastings, at around midday after reports of a collision between a car and a cyclist.
A man in his 50s has been taken to hospital by air ambulance and the road has been closed in both directions.
A police spokesman has urged anyone who witnessed the incident or who has relevant dashcam footage to email collision.apeal@sussex.police.uk, quoting Operation Xenon.
